What Is Selank?
Selank Research Peptide is a synthetic linear heptapeptide with the sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro, abbreviated TKPRPGP. It was developed as an analogue of tuftsin, an endogenous immunoregulatory tetrapeptide consisting of Thr-Lys-Pro-Arg. The additional Pro-Gly-Pro sequence was incorporated to increase resistance to enzymatic degradation. Selank is primarily investigated in neurochemical, stress-response, cognitive and immune-signalling research.

GABAergic and Peptidergic Mechanisms
Selank does not have a conclusively established high-affinity receptor. Experimental findings instead suggest indirect modulation of the GABAergic system, which provides the principal inhibitory signalling network in the mammalian brain.
GABA type A receptors are ligand-gated chloride channels. When GABA activates these receptors, chloride movement generally reduces neuronal excitability. Binding experiments indicate that Selank can alter radiolabelled GABA interactions with GABA_A receptors in a manner consistent with positive allosteric modulation. An allosteric modulator changes how a receptor responds to its natural ligand without necessarily occupying the ligand’s primary binding site. The precise Selank-binding location and receptor-subunit preference remain unknown.
Human neuroblastoma-cell experiments found that Selank alone did not directly alter messenger-RNA levels for the examined GABAergic genes. However, gene-expression responses produced by GABA changed when Selank was present, supporting an interaction with GABA-dependent signalling rather than direct transcriptional activation of those genes. Rat-brain transcriptome studies also reported changes involving neurotransmission, ion transport and synaptic-plasticity genes.
Selank has additionally inhibited enzymes that degrade enkephalins in human-serum experiments. Enkephalins are endogenous opioid peptides involved in stress, pain and emotional signalling. Slower enzymatic breakdown could prolong their activity, although the micromolar concentrations used in vitro do not establish the importance of this mechanism in living humans.
Cellular, Animal and Human Evidence
Animal studies have associated Selank with altered anxiety-related behaviour, learning, memory and brain-derived neurotrophic factor levels. BDNF supports neuronal survival, synaptic adaptation and memory-related plasticity, but changes observed in rat hippocampus and prefrontal cortex cannot confirm corresponding human outcomes.
Human evidence remains limited. One randomized Russian study included 62 participants with generalized anxiety disorder or neurasthenia and reported changes in anxiety measures and leucine-enkephalin degradation. Another comparative study evaluated Selank against phenazepam in 60 participants. These small studies lack broad international replication and do not establish efficacy across other populations or research formulations.
